Total stake: the number that matters
Whatever controls a game offers, the figure that matters is the total stake per spin, the amount that leaves your balance when you press spin. Most modern games show it clearly, often as “Bet” or “Total bet”, and many let you set it with plus and minus buttons and nothing else.

Coin-based stakes
Older and some current games break the stake into parts.

A worked example
A 20-line game with a coin value of 5p and 1 coin per line:
5p × 1 × 20 = £1.00 per spin
Raise the bet level to 2 coins per line:
5p × 2 × 20 = £2.00 per spin
Or keep 1 coin per line and raise the coin value to 10p:
10p × 1 × 20 = £2.00 per spin
Both routes give the same total stake, and in most games the same prizes. The paytable may show prizes in coins, so a “100 coin” win at a 5p coin value is £5.
Ways games
Ways to win and Megaways games have no lines to choose, so the stake is usually set directly or as coin value × a fixed multiplier. A 243-ways game might describe its stake as “25 coins” or similar; the total stake display is the reliable figure.
Adjustable lines and stake
On games with adjustable paylines, playing fewer lines lowers the total stake but means wins on inactive lines do not pay. Some games also make certain features depend on all lines being active. Check before reducing lines.

What your stake changes, and what it does not
Raising your stake makes wins and losses bigger in proportion. In nearly every slot it does not change the probability of any outcome. Some jackpot games scale the chance of triggering a jackpot with stake, and say so in their rules, but the base game is unaffected.
What it does change is your expected loss, because that scales with the total you stake. With an illustrative 96% RTP:

Choosing a stake
There is no stake that improves your chances, so the only sensible basis for choosing one is your budget. A rough approach: decide your session budget, then pick a stake small enough that a normal losing run will not use it up in a few minutes. On a high volatility game, that means a lower stake than you might expect.
